Baclofen is a gamma- amino butyric acid (GABA) agonist used as a skeletal muscle relaxant used for the relief of painful and uncomfortable muscle spasms caused by a variety of conditions.
PHARMACOKINETICS
Absorption : rapidly and almost completely absorbed from the gastrointestinal tract. Absorption may be dose-dependent, being reduced with increased doses.
Volume of distribution: 59 liters
Protein binding : 30%
Metabolism : approximately 15% of the dose is metabolized in the liver, mainly by deamination.
Elimination : rapidly and extensively eliminated from the body; excreted mainly by the kidney as unchanged drug
Half life : approximately 5.5 hours
Total systemic clearance : 180 mL/min
Renal clearance : 103 mL/min

PHARMACODYNAMICS
Eubac block polysynaptic afferent pathways & to a lesser extent monosynaptic afferent pathways (spinal cord) which inhibits the transmission of impulses through these pathways by acting as an inhibitory neurotransmitter itself. Furthermore it inhibits the release of excitatory neurotransmitters (Glutamate and Aspartate), which normally contributes to pain & spasticity.
